Things to do in Delos

Delos is a UNESCO-listed archaeological site with no accommodation and no permanent residents, so it cannot be treated as a stop on a Santorini itinerary the way Fira or Oia can.

The only way in is a boat excursion from Mykonos, a crossing of roughly 30 to 40 minutes each way, and the site closes on Mondays for much of the year, so check the current schedule before booking anything. Entry runs from about 12 to 20 EUR depending on season and guide.

A visit from Santorini would mean a ferry to Mykonos, then a separate excursion boat, then the return journey, all inside one day, which is more transit than most travellers want. This page assumes you are already based in Mykonos and deciding whether Delos fits your plans there.
